Elucidation of the molecular mechanisms regulating the deubiquitylase AMSH and its targets

Ubiquitin- and ESCRT (Endosomal Sorting Complex Required for Transport)-mediated endocytic degradation is one of the key processes in the posttranslational control of plasma membrane proteins that are important for most signal transduction pathways. The first aim of the project is to elucidate molecular mechanisms regulating the endosomal degradation pathway with a focus on the deubiquitylase AMSH (Associated Molecule with the SH3-domain of STAM) and its interactor ALIX (ALg2-Interacting protein X) in Arabidopsis thaliana. Secondly, we aim to identify AMSH-targets by proteomic analysis. The results obtained in the project are expected to contribute to the understanding of the regulation and targets of DUBs in the endocytic protein degradation.
