Architecture and structural dynamics of core components of the translational assembly scaffold

The project investigates the structural composition of core complexes involved in early to medium steps of ribosome biogenesis using novel mass spectrometric and proteomic approaches together with integrated modelling. To this end, crosslinking coupled to mass spectrometry (CX-MS) will be applied to affinity purified protein complexes from cells in order to attain a high-resolution interaction network of non-ribosomal factors involved in ribosome biogenesis. Using CX-MS, integrated modeling and EM, a molecular model of a ribosomal assembly intermediate will be generated. This approach will then be extended to include a selected subset of additional assembly factors that are required for the intermediate stages of ribosomal assembly, for which limited structural information does exist.
